Description of the research topic:

Osteogenic differentiation is a key event in bone and tooth development. Excess iron has a detrimental role in bone physiology resulting osteopenia, osteoporosis and bone fractures in patients with different forms of iron overload diseases. Recently we investigated the osteogenic differentiation potential of mesenchymal stem cells (MSCs), and found that iron inhibited osteogenic differentiation of MSCs. We lack information about whether patients with iron overload diseases have abnormalities of tooth development.
Dental pulp cells are stem cells, with a strong potential towards osteogenic differentiation, having an important role in tooth development and proper extracellular matrix formation. Under this project we will investigate the effect of excess iron on osteogenic differentiation of dental pulp cells. We will investigate how modulation of iron homeostasis influences osteogenic differentiation and extracellular matrix mineralization of dental pulp cells. We will determine mRNA and protein expressions of certain osteoblast-specific markers, such as RUNX2, SOX9, alkaline phosphatase, osteocalcin, etc. We will also measure matrix mineralization with different technics. Numerous signaling mechanisms are involved in osteogenic differentiation of MSCs. We will investigate whether the same mechanisms are implicated in osteogenic differentiation of dental pulp cells and whether excess of iron impair those signaling pathways.
