Description of the research topic:
Worldwide consent data indicate that one third of the patients with DLBCL, the most common subtype of lymphomas, cannot be cured. This study aims at a complex pathomorphological, phenotypical, cytogenetic, expression profile, signal transduction mutational as well as clinical follow up investigations as follow.
1. Retrospective analysis of 300 patients’ samples with DLBCL-NOS, DLBCL/BL and BL will take place on FFPE specimens by IH for phenotypical stratification and determination of proliferation capacity using CD5, CD10, BCL2, BCL6, MUM1, C-MYC and MIB1 markers, respectively.
2. FFPE slides of the study cohort will be investigated for C-MYC – t(8;14), BCL2 – t(14;18) BCL6 – 3q27 translocations and breaks using iFISH technology.
3. DNA will be extracted from the FFPE tissue blocks to analyse mutations in large series of signal transduction pathways using NGS in collaboration with appropriate molecular pathology laboratory.
4. RNA will be also extracted from the FFPE tissue blocks for expressions profiling using either NanoString technology and Pan Cancer Progression / Signal Pathway platforms or by sense / antisense NGS, in collaboration.
5. Homogenous disease groups will be created according to the combined IH, iFISH and expression profile analyses. The clinical staging, IPI score, EFS and OS data will be compared in these groups for prognostic stratification.
This project include and can absorb two PhD works: one related to the pathological - molecular pathological aspects, another one to the clinical aspects of these investigations.
The project has the OGYÉI/TUKEB Ethical Committee permission (50268-8/2017) in Hungary.
