Description of the research topic:
The candidate is going to study the changes of autophagic processes in ischemic/reperfused (I/R) myocardium obtained from diabetic rats under in vitro conditions. Different autophagy-related proteins including LC3II, Beclin-1, Atg-5, Atg-7, and Atg-12 will be detected. Based on preliminary results we expect differences in the autophagic processes between the nondiabetic and diabetic myocardium, however, further experiments have to be carried out to precisely understand the processes in diabetic hearts. We intend also to study the degree of apoptosis via TUNEL assay and measure the level of caspese activities. These experiments are important to understand whether there is or no difference in autophagic processes in nondiabetic and diabetic myocardium. Cardiac function and infarct size will be also registered in isolated hearts. Moreover, the incidence of ventricular arrhythmias including ventricular tachycardia and ventricular fibrillation a blood glucose will be also detected in nondiabetic and diabetic subjects. Heme oxygenase (HO-1) protein levels also will be measured.
