Blood is carried by a network of vessels lined by a single layer of endothelial cells. The vascular endothelium serves as a selective barrier to fluid and solute flux across the blood vessel walls and regulates vascular functions like barrier, inflammation, blood flow, angiogenesis and metabolism.
Reversible protein phosphorylation is one of the essential posttranslational modifications catalyzed by protein phosphatases and kinases. In the present proposal we plan a throughout investigation of the phosphorylation/dephosphorylation of angiogenesis related proteins, like thrombospondin-1 (TSP-1) protein mainly in endothelial cell model systems. TSP-1 has been shown to play crucial role in many of cellular functions via its interacting protein partners. Our goal is to clarify the effect of reversible phosphorylation of TSP-1 on its interactions, and eventually, on its physiological activities.
The results of the research can provide further information on TSP-1 related signaling pathways in normal and cancerous tissues. The results can open up novel perspectives through newly identified signaling mechanisms in vascular function regulation.
