Description of the research topic:

Although insufficient zinc (Zn) supply is a frequent problem in crop production, the molecular processes of plant responses to Zn deficiency are little known. The redox state of plant cells is regulated by the interactive signalling of reactive oxygen, nitrogen and sulfur species (ROS, RNS and RSS), but changes in their metabolism and signal interplay due to Zn shortage are unknown. Our research aims to study the changes primarily in RNS metabolism and signal transduction induced by low Zn supply in crops with different tolerance as well as in model plants. The RNS (nitric oxide, NO) -dependent signalling is examined at the level of macromolecule modifications (protein nitration, S-nitrosation, lipid nitration, RNA nitration) and of gene expression. We also intend to study whether ethylene as a frequent hormonal regulator of element deficiencies is involved in Zn deficiency response and whether ethylene signalling is related to NO signalling. A further aim of the project is to examine the possible reclamation of Zn-deficient status with zinc oxide nanoparticles or NO-releasing nanoparticles Furthermore, our aim is to identify differences and similarities in the transcriptomes of Zn-deficient and phosphorus supplementation-induced Zn-deficient Arabidopsis thaliana as well as to determine NO-dependent transcriptomic changes in NO NP-treated plants by RNA-Seq.
