Description of the research topic:

Besides the several well-known methods for enzyme immobilization new selective immobilization methods have emerged over the last decades via specific interactions of recombinant specifically tagged enzymes. The aim of PhD research is selective immobilization of His-tagged enzymes based on the immobilized metal ion affinity chromatography (IMAC). It is a well-known protein purification method but can be used as an immobilization technique also. Combining the selective complexation with other enzyme immobilization strategies can produce much more stable immobilized biocatalysts. To stabilize selectively complexed biocatalysts bifunctional supports or embedding techniques can be used. Important to compare the immobilization process with and without preliminary protein purification. Characterization of the immobilized biocatalysts: selectivity and capacity of the support to the target enzyme (SDS-PAGE, protein concentration measurement by UV-VIS and Bradford method), biocatalytic activity in enantioselective biotransformations (UV-VIS, HPLC, GC), washing resistance (against complexing agents like imidazole or EDTA), reusability. The main focus of the research is the development of efficient selective immobilization methods without separate purification step for industrially relevant enzymes.
