Description of the research topic:
Although autophagy is associated with the process, the exact autophagy processes responsible for the removal and recycling are unknown. The PhD topic aims the investigation of the localisation and local concentration of Fe and Mn in mesophyll cells, using Arabidopsis thaliana lines, affected in autophagosome formation, autophagy-dependent vacuolar chlorophagy, dark induced autophagy, chloroplast vesiculation, and senescence-associated chloroplast degradation processes. Fe and Mn locations are planned to be studies by microscopy scale particle induced and low energy X-ray fluorescence imaging, and neutron activation based techniques, combined with TEM analysis. Decomposing of the photosynthetic apparatus will be validated by BN-PAGE and mass spectrometry. Metal content below the detection limit of imaging analytic techniques will be analyzed in isolated vacuoles. The transcriptome will be analysed and assigned to the intracellular metal distribution in mesophyll cells.
